Moderna’s stock suffered a sharp reversal after data from a pivotal cancer-vaccine trial emerged online ahead of its scheduled presentation, highlighting how elevated expectations can overshadow encouraging medical results in biotechnology markets. The sell-off followed a powerful rally in the company’s shares earlier in 2026, as investors increasingly viewed its personalized mRNA cancer treatment as a potential source of growth beyond COVID-19 vaccines.

A Data Leak Triggers a Sharp Market Reaction

The reported results for intismeran autogene, Moderna’s experimental personalized melanoma vaccine developed in partnership with Merck, appeared on the medical news website Oncology Central before being removed. The full findings had been scheduled for presentation at the European Society for Medical Oncology congress in Madrid on October 24.

The apparent premature disclosure unsettled investors and contributed to a steep decline in Moderna shares on October 22. At the height of the sell-off, the stock fell more than 25%, erasing approximately $24 billion in market value. Merck shares also declined, although their losses were more limited.

The reaction illustrated the risks associated with a biotechnology stock whose valuation has become increasingly dependent on a major clinical catalyst. Moderna’s shares had climbed more than 500% during the year, leaving investors with substantial expectations for the vaccine’s performance and its potential commercial contribution.

Encouraging Clinical Results, but Expectations Were High

The Phase 3 INTerpath-001 trial involved 1,137 patients with high-risk melanoma. According to the reported findings, combining intismeran autogene with Merck’s immunotherapy Keytruda reduced the risk of cancer recurrence or death by 42% compared with Keytruda alone. The combination also reduced the risk of distant metastasis or death by 38%.

At the two-year mark, approximately 82.2% of patients receiving the combination remained alive and cancer-free, compared with 70% of patients treated with Keytruda alone.

Despite these results, investors appeared to be looking for evidence that the treatment could deliver an even greater benefit or support a broader oncology strategy. The distinction matters because a promising clinical outcome does not automatically translate into commercial success. Treatment eligibility, regulatory approval, pricing, manufacturing requirements and adoption by physicians will all influence the vaccine’s eventual financial contribution.

Moderna’s Long-Term Strategy Faces a Critical Test

The vaccine is designed around the genetic characteristics of an individual patient’s tumor, with the aim of training the immune system to recognize and attack cancer cells that may remain after surgery. Its development represents a potential expansion of mRNA technology into cancer treatment, a field that could offer Moderna new opportunities as its COVID-19-related business evolves.

However, the results do not establish that the approach will work equally well across other cancer types. Overall-survival data also remain immature, leaving important questions about the treatment’s long-term benefits.

Investors will now be watching the full conference presentation, additional clinical follow-up and the companies’ discussions with regulators. The next phase will determine whether the encouraging melanoma findings can support approval and a commercially meaningful product, or whether the market’s earlier optimism had already priced in more than the available evidence could deliver.
