Moderna’s stock experienced one of its most dramatic moves ever after the company announced positive results from a late-stage trial of its personalized cancer vaccine developed with Merck. The treatment helped reduce the recurrence of melanoma, providing investors with evidence that Moderna’s mRNA technology could have applications well beyond infectious diseases.

Shares climbed as much as 160% on Wednesday, adding to an already powerful 2026 rally. Moderna had gained 114% before the latest announcement as investors increasingly focused on its flu vaccine and broader pipeline as potential sources of future growth.

The reaction reflects a significant shift in how the market is assessing Moderna. The company has spent several years dealing with the collapse in demand for COVID-19 vaccines, which contributed to four consecutive years of stock declines and left shares almost 94% below their 2021 peak.

Short Sellers Take a $5 Billion Hit

The magnitude of the rally created an equally dramatic reversal for investors positioned against the stock. According to S3 Partners, the surge produced approximately $5 billion in paper losses for Moderna short sellers and pushed their mark-to-market losses for the year above $7 billion.

Matthew Unterman of S3 Partners described the move as exceptionally painful for short sellers and said it materially changes the risk-reward profile for investors maintaining bearish positions.

The pressure could intensify because short interest had previously reached approximately 20% of Moderna’s float before declining to around 14%. Roughly 20 million shares, or about one-quarter of short positions, had already been covered during 2026. That means a significant bearish position remained even after part of the trade had been unwound.

Could Oncology Become Moderna’s Next Growth Engine?

The most important implication of the rally is not the short squeeze itself but the possibility that Moderna’s cancer program could change the company’s longer-term earnings trajectory. Analysts reacted positively to the trial, with Needham’s Joseph Stringer describing the results as a landmark win and William Blair’s Myles Minter upgrading Moderna to Outperform.

For investors, the key question now is whether the melanoma results can translate into a commercially meaningful oncology franchise and reduce Moderna’s dependence on COVID-related revenue. The stock’s extraordinary move also means expectations have risen sharply, increasing the potential for volatility as additional clinical, regulatory and commercial milestones emerge. Going forward, investors will be watching the development of Moderna’s personalized cancer vaccine, progress across its broader pipeline and whether the company’s new oncology opportunity can justify the dramatic repricing while forcing remaining short sellers to reassess their positions.
